【摘要】:Whendeterminingthelimitingdetectableconcentrationofafluorescentsubstance,itisusuallynecessarytoincreasethereadoutscaleofaphotoelectricinstrumenttoapointwherenoise(thatis,randomfluctuationsofthesystem)becomesapparent.Thisnoisewillbesuperimposeduponthesignalfromthesample.Inmolecularfluorescencespectroscopy,thelimitofdetectionforthesamplewillbedeterminedbythelimitingsignal-to-noiseratio,S/N,wherethesignal,S,isthedifferencebetweenreadingsobtainedwiththesampleandblanksolutions,andNisthetotalroot-mean-square(rms)noise.Thelimitofdetectionforthesamplewillbegivenbytheinstrumentreadingsthatgiveasignalequaltothreetimesthermsvalueofthenoise.Note28212;Factorsotherthannoiseaffectingthesampleconcentrationcorrespondingtothelimitofdetectioninclude:thespectralbandwidthsoftheexcitationandemissionmonochromators,theintensityoftheexcitinglightthatcanbeconcentratedonthesample,thefractionofthefluorescencecollectedbythedetectionsystem,theresponsetimeofthedetectionsystem,andthepurityofthesolvent.Thesizeandarrangementofthesamplecontainerwithrespecttothelightbeamsarealsoimportant,astheyaffectboththedesiredsignalandtheextraneoussignalthatonlycontributesnoise.1.1Thistestmethodemploysthesignal-to-noiseratiotodeterminethesensitivityofafluorescencemeasuringsystemintestingforthelimitofdetection(LOD)ofquininesulfatedihydrateinsolution.Theresultsobtainedwithquininesulfatedihydrateinsolutionaresuitableforspecifyinginstrumentperformanceonsampleshavingexcitationandfluorescencebandswiderthan10nmatornearroomtemperature.1.1.1Thistestmethodisnotintendedtobeusedas(1)arigoroustestofperformanceofinstrumentation,or(2),tointercomparethequantitativeperformanceofinstrumentsofdifferentdesign.IntercomparisonoftheLODbetweeninstrumentsiscommonlyexpressedastheratioofthewaterRamanpeakintensitytotheroot-mean-square(rms)noiseasmeasuredonafluorometerusinganexcitationwavelengthof350nmThistestmethodusestheexcitationandemissionpeakwavelengthsforquininesulfatedihydrateinsolution,whichareapproximately350nmand450nm,respectively..1.2Thistestmethodhasbeenappliedtofluorescence-measuringsystemsutilizingnon-laser,low-energyexcitationsources.Thereisnoassurancethatextremelyintenseilluminationwillnotcausephotodecompositionofthecompoundsuggestedinthistestmethod.Forthisreason,itisrecommendedthatthistestmethodnotbeindiscriminatelyemployedwithhighintensitylightsources.Thistestmethodisnotintendedtodetermineminimumdetectableamountsofothermaterials.Ifthistestmethodisextendedtoemployotherchemicalsubstances,theusershouldbeawareofthepossibilitythattheseothersubstancesmayundergodecompositionoradsorptionontocontainers.1.3AtypicalLODforconventionalfluorometersusingthistestmethodis1ngofquininesulfatepermL.1.4Thesuggestedshelflifeofa1mg/mLstocksolutionofquininesulfatedihydrateisthreemonths,whenstoredinthedarkinastopperedglassbottle.1.5ThevaluesstatedinSIunitsaretoberegardedasstandard.Nootherunitsofmeasurementareincludedinthisstandard.1.6Thisstandarddoesnotpurporttoaddressallofthesafetyproblems,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriates......
